Modupe Otukoya was born on November 18, 1937, at Ijebu Imusin. She attended Women’s Training College, Ilesha, from January 1955 to December 1956. Between January 1959 and December 1960, she studied at the Provincial Teacher Training College, Osogbo. She proceeded to study at Blackburn Technical College, Lancashire, England, during the 1963/1964 academic session. From October 1968, she was at Holborn and Tothing College of Law and Arts, London until June 1971 when she graduated with LLB Law.  She attended the Nigerian Law School and was called to bar in 1973. She returned to study at the University of Lagos between 1974 and 1975.

Between 1973 and 1981, she did her private legal practice with Messrs Olajide Oyemole & Co. From October 1981 to June 1988, she was a Law Lecturer in the Nigerian Law School. In 1988, she set up the Modupe Otukoya & Co. Legal Practitioners and Notary Public, a firm which exists till date.

From 1990 to 1993, she served as Administrator of the Continuing Legal Education Programme for Legal Practitioners. She organised seminars on Continuing Education in Lagos in December 1990 and March, 1991; Enugu in May 1991; Ota in June 1992 and January 1993; and Benin in September 1992 and edited the published seminar notes.
